"summary_nl": " We want to theoretically investigate the physical behaviour of fluorescent, photoactive biosystems, in particular the interplay between their micorscopic structure and spectral functions. Understanding the nature of the excitations in photobiological systems is important form a fundamental point of view and provides a unique route to unravel phenomena such as photobleaching and blinking behavior of autofluorescent proteins, which currently limit many single-molecule techniques. Moreover, it can have an impact on biotechnology, guiding for instance the design of fluorophores with novel spectroscopic properties by selective mutations. Progress in understanding the photophysics of these systems has until now been due mainly to experimental studies: the computation of excitation energies is proving difficult, and most theoretical approaches employed so far do not possess a sufficient degree of reliability to be used as a predictive tool for the study of photoactive biomolecules. We intent to use a different theoretical framework, based on quantum Monte Carlo methods, which is currently being developed in my group for the study of photosensitive macromolecules. Armed with this tool, we want to elucidate the nature of photoexcited states of green and red fluorescent proteins, understand how mutations shift their optical transitions and investigate how dark states affect their photostability.",
